The Estimation of Phosphatase in Yeast

چکیده
For the determination of the phosphatase content of yeast many methods have been used (l-3). The usual method is to hydrolyze a glycerophosphate and measure the amount of inorganic phosphate liberated. King and Armstrong (4) have shown, however, that phenyl phosphate could be used as a substrate and the phenol det, ermined. To be satisfactory the method chosen must give a measurable amount of hydrolysis in as short a time as possible and must not require too much yeast. The advantages in using only a small amount of yeast are that there may be only limited amounts available and it reduces the amount,s of undesirable impurities frequently present. The method finally chosen was to measure the phenol liberated from a mixture of 10 mg. of yeast and 10 cc. of 0.01 M disodium phenyl phosphate, buffered with acet.ate veronal at pH 4.1, after 20 minutes at 37”. A summary of the experiments on which these choices are based follows.

The potential use of dietary probiotics to enhance the immunity and health of aquatic animals has recently attracted intensive attention. The purpose of this study was to investigate the effect of different levels of Saccharomyces cerevisiae yeast on blood serum enzymes (aspartate aminotransferase (AST), alanine aminotransferase (ALT) and alkaline phosphatase (ALP)) and antioxidant systems (S...

Leishmaniasis is a major infectious disease of considerable public health in more than 86 countries around the world. Several approaches toward vaccine development against this disease have been taken. Glycoprotein (gp63) is conserved among diverse species of Leishmania and has induced immunological responses in murine models. Therefore, this glycoprotein has been considered as a second generat...
متن کاملThe yeast phosphotyrosyl phosphatase activator is part of the Tap42-phosphatase complexes.
Phosphotyrosyl phosphatase activator PTPA is a type 2A phosphatase regulatory protein that possesses an ability to stimulate the phosphotyrosyl phosphatase activity of PP2A in vitro. In yeast Saccharomyces cerevisiae, PTPA is encoded by two related genes, RRD1 and RRD2, whose products are 38 and 37% identical, respectively, to the mammalian PTPA. Inactivation of either gene renders yeast cells ...
